Abstract

Low temperature absorption spectra were measured on chlorocruoromyoglobin reconstituted from sperm whale a pomyoglobin and chlorocruoroheme. In the electronic ground state of ferric chlorocruoromyoglobin, the spectrum of an acidic form was of a high-spin type and the spectra of an alkaline form, azide, imidazole and cyanide complexes were of a low-spin type. Complex effects of buffer species, during freezing and thawing, on the spectra of the acidic form and the azide complex in phosphate buffer and of the alkaline form in Tris-HCl buffer (pH 9) were found and their causes were discussed.
