Abstract

Recently we have determined structures of the proton coupled peptide (SLC15) and folate (SLC46) transporters using cryo-EM. Our analysis of these transporters, which builds on previous structural, biochemical and electrophysiology studies reveals important mechanistic insights into how proton binding and release drives transport in eukaryotic cells. Here we will present our current models for how proton binding modulates ligand affinity within these transporters, regulates kinetics and thermodynamically drives transport of key metabolites into the cell.
