Abstract
Some Enzymic Properties of Mitochondrial Propionyl Carboxylase
Highlights
Postulated “that when nucleotide is bound to the enzyme, as in the intermediate adenosine triphosphate (ATP)-biotin-enzyme, inhibition by avidin involves its binding by the biotin and by the adenine nucleotide.”
In no case was Ci4-propionyl pantetheine or Cl4-methylmalonyl pantetheine formed enzymically, indicating that propionyl carboxylase does not behave as propionyl or methylmalonyl-transacylase
Propionyl carboxylase purified from bovine liver mitochondria has a sedimentation coefficient (~20,W) of 19.0 S and comprises approximately
Summary
Materials and Methods-With the following exceptions the sources of all other materials were as previously described [1,2]. Extraction and Aging--An acetone powder extract is prepared by extracting 40 g of dry acetone powder with 800 ml of 0.004 M Tris, pH 7.2, for 30 minutes with occasional stirring, centrifuging at 12,800 x g for 10 minutes, and retaining the supernatant solution, which contains 6 to 8 mg of protein per ml. This solution is aged at 30” in a constant temperature bath for 20 hours with several drops of tolucne added to minimize microbial action.
