Abstract
Single nucleotide polymorphism (SNP) detection is one of the most critical genomic technologies. The SNP detection techniques today mostly utilized fluorescent dye labeled DNA probe or molecular beacon, which need high reagent cost. In this research, a genotyping system is developed by integrating miniaturized the dynamic allele-specific hybridization (DASH) technique with a bead-based microfluidic device. Our system offers a rapid genotyping scheme by using the silica microbeads conjugated to probe-gene target duplex, and performing the melting curve analysis of the duplex. Ataxia-telangiectasia mutation (ATM) gene in Landrace sows, which had been proven as a major bio-marker associated with reproductive performance, was used to demonstrate the practical applications of our genotyping system in SNP detection.
