Abstract

A simple and rapid method has been developed for the simultaneous determination of gizzerosine (GZ) and histamine (Him) in chicken tissues by HPLC using an on-column fluorescence derivatization and column-switching technique. GZ and Him in chicken tissues were extracted with perchloric acid solution, and the crude extract was purified on a Bakerbond COOH® cartridge. GZ and Him were derivatized to the respective o-phthalaldehyde fluorophore by on-column reaction, followed by column-switching and subsequent chromatographic separation on a reversed-phase C18 polymer column with gradient elution. The calibration curves were rectilinear in the range of 0.02-1μg/mL for GZ, and 0.004-0.2μg/mL for Him. The average recoveries were greater than 97% for both GZ and Him added to chicken gizzard at a level of 1μg/g.
