Abstract
Sialic acid levels in human and rabbit bone were investigated. Sialic acid was extracted from specimens of cortical bone with EDTA after grinding the defatted and dried specimens to a fine powder. After dialysis, sialic acid content was determined by the Warren assay. Thirty-five specimens of human cortical bone of various ages from fetus to 84 yr were tested. In addition, long bones from newborn to age 12-mo rabbits were analyzed, as well as callus from rabbit tibial fractures. Values in human cortical bone were 0.5-1.3 μm/g, with a mean of 0.9 μm/g. Mean values for males and females were the same, and there was no significant change with age from 4 to 84 yr. Rabbit long bone values were 0.5-1.6 μm/g. Values for human fetal and newborn rabbit long bones were 2.6 and 2.7 μm/g, respectively. Rabbit callus yielded values of 2.5 μm/g. No free sialic acid was found and no residual sialic acid could be detected after extraction.
