Abstract

IntroductionResveratrol has been indicated to promote the osteogenic differentiation of several types of mesenchymal stem cells (MSCs), such as gingival MSCs and bone marrow MSCs. Human amniotic epithelial cells (hAECs) possess the characteristics of stem cells and are showing great potential in regenerative medicine. However, whether resveratrol can be beneficial to promote the osteogenesis of hAECs has not been investigated. The current study aims to reveal the role of resveratrol in the osteogenic differentiation of hAECs and the underlying mechanism. MethodsAlkaline phosphatase (ALP) staining, alizarin red staining, the quantitative analysis of ALP activity and mineralisation, quantitative real-time PCR, western blotting, and small-interfering RNA technique were employed in this study. ResultsResults showed that resveratrol increased ALP activity and mineralisation during osteogenic differentiation of hAECs in a dose-dependent manner. Also, resveratrol significantly promoted the mRNA expression of osteogenesis-associated genes, including Runx2, ALP, Collagen-I, BMP-2, osterix, and osteopontin dose-dependently. Resveratrol also induced the protein levels of Sirt-1 and PPAR-α and activated Sirt-1 signalling. Furthermore, knockdown of Sirt-1 by RNAi significantly suppressed Sirt-1/PPAR-α signalling and inhibited resveratrol-induced mRNA elevation of osteogenesis-associated genes. ConclusionsCollectively, this study for the first time confirmed the promotive role of resveratrol during osteogenic differentiation of hAECs via Sirt-1/PPAR-α.
