Abstract

No repair of DNA single strand breaks induced by N-methyl–N-nitrosourea /MNU/ was observed in dividing meristematic tissues of barley seeds and free embryos. At O h, 3 h and 6 h after the MNU exposure, the unrepaired DNA template of free embryos inhibited both the initiation of replicons and the elongation of newly synthesized DNA pieces. Duplexes with these pieces contained more single stranded regions tnan those formed on undamaged template. Repair of all these abnormalities of daughter DNA was observed during the subsequent postreplication incubation period of 16 h-i.e. during the period of 1-2 cell cycles.
