Abstract

1. 1. There are five discrepancies between the two published sequences, those from protein and DNA, of soybean leghemoglobin a. We isolated all the tryptic peptides of leghemoglobin a and sequenced two peptides including the descrepancies. 2. 2. Our sequence agreed completely with that proposed from DNA sequencing. 3. 3. The autoxidation rate of oxyleghemoglobin a was examined in 0.1 M buffer at 25°C over pH range 5.2–7.2. 4. 4. Soybean oxyleghemoglobin a was fairly stable, the autoxidation rate being 2–3 times slower than that of mammalian oxymyoglobin over the pH range studied. 5. 5. On the other hand, it seemed that a linear relationship exists between autoxidation rate (k obs) and oxygen dissociation rate (k off) in soybean leghemoglobin, sperm whale myoglobin, yellowfin tuna myoglobin and Aplysia myoglobin.
