Purification and Characterization of Trehalase from the Yeast Arxula adeninivorans

Abstract

The yeast Arxula adeninivorans SBUG 724 during growth secretes trehalase (α,α-trehalase glucohydrolase, EC 3.2.1.28) into the liquid culture medium. Besides the extracellular enzyme internal trehalase was found and could be separated into a cell wall-bound and a soluble intracellular fraction. Investigating the enzymes we show that their properties were similar. The optimum temperature for the enzyme activity was 45 to 55°C and the highest activity was obtained in a p H range between 4.5 and 4.9. The enzymes were specific for trehalose and the K m values were determined to be 0.8–1.0 mM. The molecular mass of the trehalases was 250 kDa. Die Hefe Arxula adeninivorans scheidet während des Wachstums Trehalase (α;α-Trehalose-Glucohydrolase, EC 3.2.1.28) in das Kulturmedium aus. Neben diesem extrazellulären Enzym konnte ein intrazelluläres und ein Zellwand-gebundenes Enzym isoliert werden. Alle drei Trehalaseaktivitäten haben ähnliche Eigenschaften. Ihr Temperaturoptimum lag zwischen 45 und 50°C und ihr p H-Optimum zwischen 4,5 und 4,9. Die Enzymaktivitäten waren spezifisch für Trehalose. Der K m -Wert lag zwischen 0,8 und 1,0 mM Trehalase. Die molekulare Masse betrug 250 kDa.
