Abstract

Extraction of soluble proteins from most animal tissues is relatively straightforward, since the cell membranes are weak and easily ruptured by a combination of mechanical and osmotic forces. The tissue is disrupted in a suitable buffer using a homogenizer and the homogenate then clarified by centrifugation. Extraction of membrane proteins requires more specialized methods, and these are dealt with in Chapter 28.
