Abstract

Objective: To verify the inhibitory effect of FSMLE, which is prepared from the fruit kernel of Tianguo, on human hepatoma HepG2 cells, and to explore its potential mechanism on human hepatoma HepG2 cells. Methods: The activity of FSMLE on human hepatoma HepG2 cells was detected by CCK8 kit; the migration ability of FSMLE to human hepatoma HepG2 cells was detected by scratch test; the apoptosis of human hepatoma HepG2 cells induced by FSMLE was detected by TUNEL kit; the expression of Bax in human hepatoma HepG2 cells by FSMLE was observed by immunofluorescence; the expression of Bax and Caspase-3 proteins in human hepatoma HepG2 cells was detected by Western Blot assay. Results: In HepG2 cells, the dose of FSMLE within the range of 20~160 μ G/ml can significantly inhibit the proliferation of HepG2 cells in a dependent manner of concentration and time, and migration of HepG2 cells; it can also induce apoptosis of HepG2 cells; and increase the protein expression level of Bax and caspase3. Conclusion: The extract of Semen FSM has obvious inhibitory effect on the proliferation of human hepatoma HepG2 cells and is closely related to the induction of apoptosis.
