Abstract
Fluorescent method is a powerful tool for genotyping point mutations. Current existing methods usually need cost labeled fluorescent probes, which causes great limitation to their applications in clinic. Here, a fidelity method for genotyping point mutations has been developed based on melting-point difference without labeled fluorescent probes. The method employs ligase to ligate two specific probes to produce a high melting temperature. The homozygotes and heterozygotes are scored accurately by obvious melting-point difference among genotypes. This method would provide an accurate and economical-cost tool for point mutation genotyping.
