Event Abstract Back to Event Phenotype and function of lymphocytes in normal and inflamed appendix Ghasem Mosayebi1* 1 Arak University of Medical Sciences, Immunology, Iran Introduction: The appendix is considered as part of the gut- associated lymphoid tissue. The function of appendix is not clearly defined. In this study to understand better the immunological function of the appendix, we investigated the function and phenotypic pattern of lymphocyte subsets in peripheral blood and appendix of patients with normal and inflamed appendix tissue. Patients and Methods: Peripheral blood samples and appendiceal mononuclear cells obtained from 80 patients (mean age; 23±10.5), who were clinically suspected of having appendicitis. Based on histopathological examination, twenty-five patients had normal appendix while 40 and 16 were diagnosed with acute and gangrenous/perforate appendicitis, respectively. The phenotypic characteristics of lymphocyte subsets in blood and tissue analyzed by three color-flow cytometry. The proliferative responses of peripheral blood or tissue mononuclear cells assayed by MTT method. Results: The frequency of CD19/DR, HLA-DR and CD19+-cells in the appendix tissue were significantly higher than the peripheral blood in all the groups (p<0.001). Overall, there were significantly higher percentage of CD5, CD3, CD8, α/β-TCR, RA/RO, CD3/RA in peripheral blood of normal patients than were found in appendix mononuclear cells (p<0.01). In patient with acute appendicitis, the percentage of CD5, CD3, CD8, α/β-TCR, CD3/CD38 and CD3/RA were significantly different between blood and tissue (p<0.001). There were significantly different in percentage of CD19, HLA-DR and CD3/CD4/αβ-TCR between acute, gangrenous appendicitis patients and patients with normal appendix. There is significant different in the proliferative responses of the peripheral blood and appendiceal mononuclear cells to PHA and Con-A between acute, gangrenous appendicitis patients and patients with normal appendix (p<0.01). Conclusion: The phenotypic and function of lymphocytes are different between peripheral blood and appendix tissue. Also, lymphocytes phenotypic changes in peripheral blood of appendicitis patients can be used as a diagnostic marker for appendicitis.
