Abstract
The release of [ 3H]5-hydroxytryptamine ([ 3H]5-HT) evoked by 15 mM KCl in superfused rat cerebellum synaptosomes was inhibited by 5-HT (pEC 30=8.73). Methiothepin antagonized 5-HT (pA 2=9.28); ketanserin, methysergide, cinanserin and spiperone were ineffective. The receptors involved were activated (pEC 30=8.90) by the 5-HT 1 agonist 5-methoxy-3-[1,2,3,6-tetrahydropyridin-4-yl]-1H-indole (RU 24969)·(−)Propranolol shifted to the right (pA 2=8.05) the dose-response curve of 5-HT. The 5-HT 1A agonist 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT) was ineffective. In conclusion, autoreceptors are present on 5-HT nerve endings in rat cerebellum and appear to belong to the 5-HT 1B subtype.
