Abstract

Digital holographic techniques in microscopy allows minimal invasive, full field quantitative phase contrast imaging of transparent biologic specimens as well as flexible and versatile configuration of wavefronts in the focus plane of an objective in order to perform pN and fN forces. Both digital holographic (DH) techniques – one based on phase shifting microscopy and one on liquid crystal on silicon (LCOS) devices – are usually applied separately. In this work, the simultaneous realization of both modes is demonstrated and optimized. The setup is characterized with different biologic specimen.
