Abstract

The success of comprehensive proteome analysis is highly dependent on the quality of the sample fractionation techniques employed prior to analysis by mass spectrometry. Novel fractionation tools like OFFGEL fractionation and gel eluted liquid fraction entrapment electrophoresis (GELFrEE) separate proteins or peptides according to their isoelectric point and molecular weight respectively, whereby the separated components are recovered in solution. Liquid-phase protein separation platform with a high degree of resolution, throughput, high sample recovery and compatibility with downstream analysis would present a more desirable and useful system. These novel tools can enrich the target proteins from any complex protein mixtures resulting in higher confidence in the identification of proteins even at low concentration. This review critically examines and outlines the findings of the recent applications of novel liquid-phase protein recovery strategies (OFFGEL and GELFrEE) and their application in proteomic studies especially in food quality, safety and authenticity research.
