Abstract

α-amanitin is a selective RNA polymerase II inhibitor. Treatment of pachytene spermatocytes with this drug leads to the fragmentation of the nucleoli. This fragmentation proceeds more slowly in the pachytene spermatocytes than in the Sertoli cells. Autoradiographic evidence indicates that the extranucleolar HnRNA synthesis is blocked in the pachytene spermatocytes by α-amanitin whereas nucleolar RNA synthesis remains unaffected. Even a completely fragmented nucleolus in a pachytene spermatocyte still synthesizes RNA. In Sertoli cells both the extranucleolar and the nucleolar RNA synthesis is blocked by α-amanitin. These results are discussed and it is suggested that rRNA synthesis in pachytene spermatocytes in some respects differs from that in somatic cells.
