Abstract

Summary This is the final article of a series of three. After briefly recapitulating the first two, this article will move on to consider the sources of malignant disease in the real world: diet and the bowel flora are important in this context. We will then look at some of the intermediate stages between carcinogenesis and the appearance of frank malignancy—here the topological order of DNA and oncogenes are important. Insight into natural resistance to carcinogenic influences can be gained from the study of congenital DNA repair syndromes. We will then look in more detail at the nature of these DNA repair systems and the ways in which it is possible to stimulate them. From such understanding effective therapy may evolve.
