Some lignans, like arctigenin posses antitumor, anti inflammatory, antiviral and antioxidant properties. Furthermore, it was also reported that arctigenin – a member of phenolic molecule family – has neuroprotective activity against glutamate-induced toxicity in primary neuronal cultures and its direct interaction with kainate receptors (KARs) [Jang et al. (2002) J Neurosci Res 68:233-40]. In the present study the effect of arctigenin, produced by plant tissue cultures or obtained commercially was tested. In vitro electrophysiological experiments were performed on cortical slices of adult rats, and changes of evoked field potentials were tested in somatosensory cortex. Alterations of the early and late components of evoked responses were evaluated. Furthermore, the effect of arctigenin on evoked field potentials was investigated in in vivo electrophysiological experiments, using anesthetized rats. [3H]kainate ligand binding studies of recombinant KARs expressed in HEK-293 cells were performed to establish the subunit selectivity of arctigenin/KAR interaction. Co2+-uptake assay was used to assess arctigenin-induced changes in non NMDA receptors mediated Ca2+ influx into neurones. Our results indicate that arctigenin dose dependently inhibited both components of the evoked response in brain slices and the same effect was observed in layer 3 evoked responses recorded from anesthetized rats. According to receptor binding studies antagonistic effect of arctigenin is partly mediated by the inhibition of KARs. Ca2+ permeability of non NMDA receptors was also dose dependently reduced by arctigenin in somatosensory cortex. Our results demonstrated the antagonistic effect of arctigenin on non NMDA receptors in rat somatosensory cortex. Since non NMDA receptors are involved in different brain diseases, a low-affinity antagonist compound can be useful in medical treatments.
