Abstract

To study the net uptake of serum protein by lymph chylomicrons in vitro, whole chyle obtained from the thoracic duct of rats fed olive oil was incubated for 30 min at 37° with rat serum from which S f > 400 lipoproteins had been removed or with one of the following fractions of that serum: total low density lipoproteins, high density lipoproteins, an albumin-rich fraction, or a globulin-rich fraction. S f > 400 lipoproteins were then isolated in a swinging-bucket rotor using a non-linear density gradient, and their protein contents determined by elemental N, C and H analysis. Distinct increments in the protein content of the isolated S f > 400 lipoproteins were observed after incubating chyle with serum, both lipoprotein fractions and the globulin-rich fractions. (The protein content of these S f > 400 lipoproteins isolated after incubating chyle with serum was similar to that of S f > 400 lipoproteins isolated from serum of fat-fed rats.) The results indicated that the major portion of the protein taken up by lymph chylomicrons was derived from lipoproteins rather than other serum proteins.
