Abstract

目的:研究不同浓度烟酸诺氟沙星对乌鳢肝微粒体细胞色素P450酶系的影响,旨在为水产养殖合理用药提供安全指导。方法:健康乌鳢60尾,体重200 ± 20.0 g,随机分为空白对照组、低剂量(5 mg/kg)、中剂量(20 mg/kg)、高剂量(40 mg/kg)烟酸诺氟沙星浓度组,每组15尾,连续口灌给药5天。采用差速离心法提取肝微粒体,利用底物代谢法联合紫外分光光度计测定NADPH-细胞色素C还原酶(NCCR)、苯胺-4-羟化酶(AH)、氨基比林-N-去甲基酶(AND)、红霉素-N-脱甲基酶(ERND) 4种酶的含量。结果试验结果表明,与对照组相比,烟酸诺氟沙星组微粒体蛋白的含量明显减少(p < 0.05),而NCCR、AH、AND、ERND4种酶的活性则显著提高(p < 0.05)。且随着药物浓度的升高,表现出活性增强的趋势。结论:烟酸诺氟沙星对乌鳢肝微粒体细胞色素P450酶系活性具有诱导作用。 The present study was conducted to evaluate the effects of niacin norfloxacin on the activity of CYP450 enzyme in liver of Channa argus. The aim of the study was to provide security guidance for rational use of niacin norfloxacin in aquaculture. C. arfus were fed with different concentrations (0 mg/ml, 5 mg/kg, 20 mg/kg, 40 mg/kg) of niacin norfloxacin by oral administration for 5 days. The activities of enzyme of NCCR, AH, AND and ERND were determined. Our results demonstrated that microsomal protein content in niacin norfloxacin groups were decreased significantly as compared with control group, and the activities of NCCR, AH, AND and ERND were significantly increased in a dose-depended model which indicated that niacin norfloxacin could promote the activity of those enzyme.
