Abstract

We present high-throughput single-molecule manipulation using a benchtop centrifuge, overcoming limitations common in other single-molecule approaches such as cost, throughput, technical difficulty, and infrastructure requirements. An inexpensive and compact Centrifuge Force Microscope adapts to a commercial centrifuge enabling use by non-specialists, and integration of DNA nanoswitches facilitates reliable measurements. We demonstrate new features by measuring temperature and magnesium dependent DNA unzipping together with multiplexed DNA force-extension and overstretching.
