Abstract

Fundamentals of multidimensional liquid chromatographic techniques is overviewed, including off-line, serial, stop-and-go, heart-cutting and comprehensive separation modes, with attention to resolution, peak capacity, efficiency, orthogonality and selectivity. Experimental setup, modulation of the fraction collection and transfer from the first to the second dimension, selection of suitable columns, flow-rate, fraction volumes and mobile phase, compatibility of mobile phases in comprehensive LC×LC, 2D asymmetry and are addressed. The survey presents various coupled two-dimensional modes (RP×RP, HILIC×RP, IEX×RP, SEC×RP and discusses possible issues connected with their on-line coupling.
