Abstract

Abstract Murine monoclonal antibodies (McAbs) which have been produced against the IgG1 allotypes G1m(z) clone 5A1, G1m(a) clone 5E7 and G1m(f) clone 5F10, were investigated further for their reactivity and specificity in different assays. In the haemagglutination inhibition assay they appeared to be good reagents for Gm-typing. However, in a direct ELISA with wells coated with myeloma proteins, some cross-reactivity was seen with some of the other IgG proteins tested. An alternative assay for the determination of Gm allotypes was set up by using a capture ELISA, in which the wells were coated with the purified monoclonal anti-G1m antibodies. After incubation with the test sample, a peroxidase-labelled monoclonal anti-IgG antibody was used. Since, in this way, a reliable typing assay for G1m(z), G1m(a) and G1m(f) was developed, it was investigated whether this assay could also be used for the quantification of these allotypes. Although the coefficients of variation of the different determinations are still high, the results obtained with these McAbs in the capture ELISA are very promising for a quantitative determination of Gm allotypes
