Abstract

Summary The uptake and metabolism of indoleacetic acid (IAA) was studied in habituated embryogenic and non-embryogenic ovular callus lines, using IAA-1-14C and IAA-2-14C. Both embryogenic and non-embryogenic callus decomposed IAA rapidly. While rate of decomposition for both callus types was similar when grown in light, embryogenic callus decomposed IAA at a higher rate in the dark. Chromatography of ethanolic extracts revealed a large peak of activity in addition to an IAA peak, corresponding to indoleacetylaspartate in the embryogenic callus. This complex was typical for embryogenic callus. It is suggested that by using this mode of IAA removal, auxin level in embryogenic callus is reduced to a level inductive to embryogenesis.
