Abstract

Ultrafast endocytosis generates vesicles from the plasma membrane within 100 ms after synaptic vesicle fusion. Dynamin is required for the fission of ultrafast endocytic vesicles. However, dynamin molecules are cytosolic and require ∼10 seconds for their recruitment and assembly. Thus, it is not clear how dynamin mediate fission of vesicles on this time scale. To reveal molecular underpinnings of this process, we coupled genetic depletion with flash-and-freeze microscopy, which visualizes membrane dynamics of synapses in electron micrographs with millisecond temporal resolution.
