Abstract

Objective To study the mechanisms of diaphragmatic muscle cell apoptosis in rat pulmonary emphysema and the mechanisms of bone marrow mesenchymal stem cells transplantation against apoptosis.Methods Female Lewis rats were randomly divided into three groups:normal control group,emphysema group,emphysema + MSCs transplantation group.Weight of diaphragmatic muscle,the content of MDA,SOD activity,cell apoptosis index(AI),the protein of Bcl - 2 and Bax were detected in each group.Results The weight of diaphragmatic muscle and SOD activity in emphysema + MSCs transplantation group and the emphysema group were less than that of the normal control group ( P < 0.01),but the content of MDA,AI and Bax were higher than that of the normal control group ( P <0.01). The weight of diaphragmatic muscle and SOD activity in emphysema + MSCs transplantation group was higher than that of the emphysema group( P < 0.01 ),but the content of MDA,AI and Bax were less than that of the emphysema group ( P < 0.01 ). Conclusions Elevated levels of oxidative stress induced diaphragmatic muscle cell apoptosis in rat pulmonary emphysema,and the transplantation of MSCs can decrease the diaphragmatic muscle cell apoptosis of pulmonary emphysema rat by reducing the level of oxidative stress.
