function, reduction of individual disulfide bonds (∆C137 or ∆C145) decreased cytokine expression by 35+6% and dual reduction decreased cytokine expression by >60% (p<0.05). Wild type PAP2 induced translocation of NF-κB into the nucleus and increased IκB phosphorylation: this was not observed with deletion of the C terminus (<2% activity) and was proportionately reduced with mutation of disulfide bonds (∆C137) (p<0.05). Conclusions: The C-terminus of PAP2 and conservation of its intact disulfide bonds activate macrophages via an NF-κB pathway. A unique 30 amino acid fragment associated with conserved 3-D conformation of the PAP protein is critical for its activity.
