Highlights

  • Use of Microsomes and Hepatocytes in Drug Discovery and Development subcellular fractions for drug metabolism studies as it is inexpensive and easy to handle [4]

  • In order to determine intrinsic clearance of new chemical entities (NCEs), which is a useful parameter for screening process of stable compounds and for establishing an in vitro correlation between animals and humans, microsomal assays play significant role

  • There are few limitations of microsomes, for instance, they have to be supplied with co-factors like NADPH, or UDPGA for the metabolic reactions to initiate, and lack the cell membranes to mimic the physiological environment in hepatocytes [7, 8]

Summary

Introduction

Use of Microsomes and Hepatocytes in Drug Discovery and Development subcellular fractions for drug metabolism studies as it is inexpensive and easy to handle [4]. Microsomal assays are the default assays for metabolism and DDI studies at the drug discovery stage [5]. This assay can be utilized to assess extra-hepatic metabolism and determine the DDI potential of the drug candidate in preclinical phase [6].
