Abstract

1. 1. Proteins were estimated by densitometry after electrophoretic separation in agar and staining with the appropriate reagents. 2. 2. Some problems concerning the optical measuring device were extensively studied (influence of free lateral diffusion; effect of the ratio of the length of the coloured zone to the length of the scanning slit), and procedures were developed for calculating the optical density curves and the areas outlined by them. 3. 3. Several preparations of proteins isolated and purified from normal human sera were examined by this method. In every case, a good linear relation was obtained between the intensity of the coloration and the protein concentration in the sample. The capacity of certain proteins to bind Amidoschwarz appears to be identical, but the results are not sufficiently conclusive to permit one to apply this statement to all serum proteins. 4. 4. The advantages of electrophoresis in agar as compared with paper electrophoresis, as far as the quantitative examination of the proteins is concerned, are discussed.
