Abstract

Objective To investigate the inhibitive effects of siRNA on the expression of β-cate-nin and hepatic cancer cellular proliferation. Methods RT-PCR analysis, MTT and western blotting were used to detect the expression of β-catenin, cellular proliferation and protein expression of p-catenin , respectively. Results The expression of β-catenin in group A and B was less than group C and D on the first day. The expression of β-catenin in group A and B was less than group C and D on the second day, and there was no expression on the third day. There was significant difference between test groups (A, B ) and controls (C and D) (P<0. 01). The inhibitive rate of cancer cellular proliferation by RNAi for group A and B was significantly higher than the controls (C and D). The protein expression of P-catenin was also inhibited by siRNA in group A and B. Conclusion RNAi is of great value in the treatment of hepatic cancer. Key words: Carcinoma hepatocellular; β-catenin; HepG2 cells; RNAi
