Abstract

Infection of Buffalo green monkey kidney cells with Hallé (subacute sclerosing panencephalitis) (SSPE) virus yields a cell-free virus titer of ∼10 8 PFU/ml and causes accumulation of masses of intracellular nucleocapsids. A simple and rapid purification method employing Renografin-76 gradients has been used to purify both virus and nucleocapsids. This cell-virus system and purification procedures have facilitated the preparation of homogeneous SSPE genomic RNA isolated by isokinetic sucrose gradients. The RNA has been used as a template for synthesis of a complementary DNA probe.
