Abstract
Legionella is a potential causative pathogen in pneumonia. It is also a facultative intracellular bac terium and, because it multiplies in macrophages, results of in vitro susceptibility tests do not always reflect clinical therapeutic effect. For this reason, it is necessary to test for both intracellular penetra tion of the agent and bactericidal activity against intracellular multiplying Legionella. In this study, the antibacterial activity of sparfloxacin, a newly developed fluoroquinolone, against Legionella was studied and compared with that of ofloxacin, ciprofloxacin, norfloxacin, and erythromycin. 1. Materials and Methods The minimum inhibitory concentrations (MICs) for the Legionella species were determined by the micro dilution broth method. A total of 68 Legionella strains were used in this study: standard reference strains of L. pneumophila (serogroups 1 to 13, n = 14) and Legionella spp. (n = 27); and clinical isolates of L. pneumophila (serogroup 1, n = 21; serogroup 2, n = 2; serogroup 3, n = 1; Table I.
