Abstract

In this study, Microsatellites were screened from genes and ESTs of Scylla paramamosain and the related species Scylla serrata. Eighteen polymorphic loci were detected, with a total of 106 alleles. The observed and expected heterozygosity per locus ranged from 0.08 to 1.00, and from 0.08 to 0.88. Polymorphism information content (PIC) showed that 12 loci were high informative (PIC > 0.5). Two loci were found to deviate significantly from Hardy–Weinberg equilibrium after Bonferroni correction (P < 0.0028). Significant linkage disequilibrium was detected between loci Scpa-ALF-SSR and Scpa-SSR03 after Bonferroni correction (P < 0.0003). Cross-species amplification of these 18 microsatellite markers showed that most of them could give the expected products in three other species: S. serrata, Scylla tranquebarica and Scylla olivacea. This study will be useful for construction of genetic linkage maps, mapping of quantitative trait loci (QTL), and analysis of comparative genome in S. paramamosain and the related taxa.
