Abstract

Human α 1-acid glycoprotein was isolated from urine proteins of a paizing rabbits. Using purified α 1-acid glycoprotein and its anti-DEAE-cellulose and CM-cellulose. The isolated preparation was found to be homogeneous by electrophoretical, immunological, and ultracentrifugal analysis, and was immunologically identical to serum α 1-acid glycoprotein. A potent antiserum specific to this protein was obtained by immunizing rabbits. Using purified α 1-acid glycoprotein and its antiserum thus prepared, the concentration of α 1-acid glycoprotein in human serum was determined by single radial immunodiffusion. As a consequence, the mean value in serum of this glycoprotein was 96.5 ± 3.1 mg/dl for normal individuals.
