Abstract

Tinospora cordifolia (wild.) Miers ex Hook. F. & Thoms is well known for its use in traditional as well as modern medicine for its application in various diseases like immunomodulatory, anti-diabetic, anti-oxidant, hepato-protective, anti-arthritic properties. The present study reports a HPTLC and molecular fingerprinting analysis of T. cordifolia plant that can be used as a reference for identification and quality control of the products. The PCR amplification with rbc La-F and rbc La-R ITS region showed 600bp PCR product. The DNA sequence analysis of samples used in this study were submitted with the NCBI nucleotide database and it confirmed that the collected samples are of T. cordifolia. The quantitative analysis of the plant material also revealed the presence of various amount of alkaloids, carbohydrates, tepenoids, steroids, phenolic compounds, glycosides, saponins, flavonoids, proteins. The HPTLC fingerprinting was performed by derivatization with Chloroform: Methanol (9: 1) mobile phase and Anisaldehyde sulphuric acid reagent. HPTLC fingerprinting profile using CAMAG HPTLC tools showed 11 bands at visible light in 10μL of methanolic stem extract and 13 bands at visible light in 10μL of methanolic callus.
