Abstract
To the Editor: Atopic dermatitis (AD) is a heritable and heterogeneous inflammatory chronic skin disorder that makes the “one size fits all” therapeutic approach suboptimal for patients.1,2 Using decision tree/supervised learning of extensive clinical, molecular, and genetic data, we aimed to define distinct AD endotypes. Deep phenotyping and whole-genome sequencing were performed on samples obtained from participants of Evaluating the Effects of Tradipitant vs. Placebo in Atopic Dermatitis (EPIONE), a randomized, placebo-controlled phase III study in patients with severe pruritus comprising mild (23%), moderate (64%), and severe (13%) AD, as determined by the validated Investigator Global Assessment (IGA) for Atopic Dermatitis scale.
