Abstract

The research on molecular biology in ramie [Boehmeria nivea(L.)Gaud.] is not as advanced as that in major crops due to the lagged development of molecular markers.We compared the effectiveness of restriction site amplified polymorphism(RSAP),sequence-related amplified polymorphism(SRAP),and simple sequence repeat(SSR)markers in revealing genetic relationship of 16 entries of ramie germplasm.The SRAP marker detected the most polymorphic loci,followed by the RSAP marker,and SSR marker had the lowest polymorphism.The dendrogram based on SRAP or RSAP data showed high consistency with that based on combined data of SRAP+RSAP+SSR,and their genetic distances were significantly correlated.In contrast,the SSR data constructed a dendrogram with different clusters of the 16 entries.The similarity of clusters between phenotypic traits and molecular markers was sequenced as SRAP+RSAP+SSRRSAPSRAPSSR.The SPAR marker had the highest efficiency to disclose the genetic relationship among ramie germplasm,which was slightly higher than that of the RSAP marker,whereas the SSR showed the lowest efficiency.Thus,RSAP marker was applicable in diversity analysis in ramie,and the joint use of RSAP,SRAP,and SSR markers may enhance the reliability of genetic relationship.
