Abstract

Objective: To develop a novel delivery of curcumin by formulating its transfersome into nasal in situ gel. Methods: Curcumin transfersomes were prepared by thin film hydration method. Optimization of the formulation was performed using Design Expert software. Formulation and characterization of optimized batch of curcumin transfersomes were performed. The nasal in situ gel was prepared by incorporating the optimized transfersome. Results: The pH, drug content, in-vitro drug release profile and thus the short-term stability study revealed that the formulation was stable during study period. The drug release mechanism was found to be non -fickian transport kind of diffusion. In-vivo study shows that the curcumin concentration in the brain after IN administration was noticeably higher than the concentration following IV administration. Conclusion: Curcumin loaded transfersomes integrated into nasal in-situ gel prolongs the drug contact time and releases the drug in a controlled manner, which results in improved bioavailability, reduced dose requirements, improved patient safety and acceptability.
