Abstract
Introduction: T lymphocytes play an important role in the etiopathogenesis of Crohn' s disease (CD). Data about intracellular cytokine profiles detected by flow cytornetry are scarce. Aim: To study Thl (IFN-'Y, IL-2) and Th2 (IL-4, IL-IO) cytokine profile in PBL,IEL and LPL in patients with fistulizing (F) and non-fistulizing (NF) CD. Patients: Thirteen patients with perianal fistulae and ten patients with non-fistulizing CD were included. Control population (C) consisted of 12 patients with a normal colonoscopy. Methods: Ten colonic biopsy specimens were obtained. IEL were isolated mechanically and LPL using collagenase type IV. PBL were isolated using Ficoll-Paque gradient. Intracytoplasmic IFN-'Y, IL-4, IL-2 and IL-1O were determined by flow cytometry after PMA-ionomycin stimulation. Results: open bars:PBL; black bars:LPL. No differences were seen for IL-4, IL-1O and colon IEL. Conclusion: A downregulation ofThl cytokine profile was observed in colon LPL in patients with F and NF CD. In PBL, the downregulation was only noted in non-fistulizing CD.
