Abstract

In order to simplify the pretreatment procedures,tobacco sample was directly extracted by methanol and the UPLC-UV conditions were optimized,then the solanesol content in the obtained extract was determined.The results showed that: 1) The optimal conditions of extraction were: methanol as solvent and extracting for 20 minutes at 25 ℃.2) The concentration of working curve of this method ranged from 10 to 200 mg/L.Good linear relationship existed between mass concentrations and peak areas with the correlation coefficient of 0.9997;3) The detection limit of this method was 0.054 mg/L with the relative standard deviation of 2.1% and the average recoveries of 105.5%.This method is fast,simple,accurate and suitable for the determination of solanesol content in tobacco.
