Abstract

In order to simplify the pretreatment of tobacco leaf samples,leaf sample was directly extracted by hexane and the obtained extract was saponified with potassium hydroxide,the solanesol concentration in saponified extract was determined by HPLC.The result showed that: 1) the efficiencies of extraction and saponification were better when the extraction lasted 24 hrs at 65 ℃ and the extract was saponified with 0.01 mol/L KOH for 0.5 hrs;2) the peak area had good linear relationship with mass concentration in the concentration range of 2.125 to 34.000 mg/L(r=0.99998);3) the average recovery was 97.6% and the relative standard deviation(RSD) was 2.7%(n=3).The method was rapid,simple and accurate.
