Expression of microRNA-141 in bladder cancer and its correlation with tumor grade

Abstract

Objective To investigate the expression of microRNA-141(miR-141) in bladder cancer and its correlation with tumor grade. Methods A total of 80 patients with bladder cancer admitted to the department of Urology of the Second Hospital of Shanxi Medical University from March 2012 to March 2014 were selected, including 67 males and 13 females, aged(68.23±3.57)years old, ranging from 61 to 83 years old.The relative expression of miR-141 mRNA in bladder cancer tissues and adjacent normal tissues were detected by real-time polymerase chain reaction(RT-PCR). Correlation of miR-141 expression with clinicopathological characteristics and prognosis of bladder cancer patients was analyzed.The overall survival rate(OS)and progression-free survival rate(PFS)were compared between patients with low miR-141 expression and high miR-141 expression. Results The relative expression of miR-141 mRNA in bladder cancer tissues was significantly higher than that in normal adjacent tissues [(4.93±0.88)versus(0.83±0.11), P 0.05). The relative expression of miR-141 mRNA had significant difference among bladder cancer patients with different tumor grades, tumor stages, muscular invasion and tumor metastasis(P<0.05). The 3-year survival rate of bladder cancer patients with different tumor grades, tumor stages, tumor metastasis, and miR-141 expression showed significant difference(P<0.05). Tumor grade, tumor stage, tumor metastasis, and miR-141 expression were independent risk factors that affect the prognosis of bladder cancer patients(P<0.05). OS and PFS of miR-141 low expression patients were significantly higher than those of miR-141 high expression patients, with statistic difference[70.97%(22/31)versus 20.41%(10/49); 64.52%(20/31)versus 24.49%(12/49), P<0.05]. Conclusion The miR-141 is highly expressed in bladder cancer patients, and is closely related to tumor grade, tumor stage, tumor metastasis and prognosis of bladder cancer patients, which can provide a reference for early diagnosis and prognosis of bladder cancer patients.
