Abstract
Objective To investigate the expression and role of Kruppel-like factor 8 (KLF8) in hepatocellular carcinoma (HCC).Methods The KLF8 mRNA and protein levels in different HCC cell lines and HCC tissues were determined by RT-PCR, immunochemistry and Western blot respectively.Resuits Metastatic potential HCC cell lines had higher KLF8 mRNA and protein levels than primary HCC cell lines (P <0.05).Relative KLF8 mRNA levels in normal liver tissues (6 cases) ,HCC tissues (17 cases), and recurrent HCC tissues (6 cases) were 0.2377 ± 0.0658, 0.4683 ± 0.2073, and 0.6669 ± 0.0239, respectively (P < 0.05).Metastasis specimens exhibited the highest level of KLF8 expression (5/6,83.3 % strong positive), and primary tumor tissue specimens (27/33,81.8%) had higher level of KLF8 expression than normal (1/6,16.7%) and liver cirrhosis tissue specimens (2/6,33.3%), (P < 0.01).Conclusion Abnormally activated KLF8 expression may contribute to HCC development and progression.
