Abstract
Objective To explore the expression of aquaporin1 and aquaporin3 in the donor lung with warm ischemia, and study whether the,expression level of aquaporinl and aquaporin3 is related to the acute lung injury caused by warm ischemia. Methods Adult Wistar rats were divided into 3 groups ac-cording to the warm ischemic time of 0 h (group A,n = 10) ,1 h (group B,n = 10) ,and 2 h (group C, n = 10). We examined the expression level of AQP1 and AQP3 protein and mRNA in the lung tissues by immunohistochemistry, Western blotting, and reverse transcriptase-polymerase chain reaction. Results Immunohistochemical examination revealed that AQP1 and AQP3 were expressed in 3 groups. The mRNA average A values of AQP1 were 0.63±0. 10,0.49±0.07, and 0.36±0. 06, and those of AQP3 were 0.85 ± 0. 06,0.76 ± 0.08, and 0. 52± 0. 07 in groups A, B, and C, respectively. The protein average A values of AQP1 were 1.13±0.18,0.88 ± 0.13, and 0.76±0.09, and those AQP3 were 1.92 ± 0. 26. 0. 95± 0. 15, and 0.61±0. 07 in groups A, B, and C, respectively. Conclusion We conclude that AQP1 and AQP3 may play an important role in the maintenance of water homeostasis and are related to water movement and acute lung injury caused by warm ischemia.
