Abstract

Herbicides which are inhibitors of photosynthetic electron transport bind to two different proteins at the reducing side of photosystem II. A 32–34 kDa protein is responsible for binding of diuron and related herbicides (e.g. atrazine, metribuzin) and photosystem II reaction center proteins (molecular weight range 41–53 kDa) are responsible for binding of phenolic herbicides (e.g. ioxynil, dinoseb). This is concluded from binding experiments of radioactively labelled herbicides of both classes to isolated chloroplasts from susceptible and s-triazine-resistant Amaranthus retroflexus, and to protease treated chloroplasts from spinach and from photoaffinity labelling experiments of spinach chloroplasts and photosystem II particles with azide-derivatives of both classes of herbicides.
