Abstract
Highly purified, 750A, dense-core vesicles from bovine splenic nerve can be prepared with a norepinephrine(NE) content of 100 nmoles/mg protein-'. Upon incubation at 30°C, however, the NE is rapidly lost from two pools with halflives of ∼ 4 and 35 minutes, even in the presence of ATP. Loss from the slower, ATP-sensitive pool accounts for 807o of the total vesicle NE and is accompanied by decrements in vesicle matrix density viewed at the ultrastructural level. It is known that the density of the matrix substance parallels semi-quantitatively the vesicle NE content under these conditions.A search was undertaken for factors which normally stabilize vesicle NE content in vivo, but are lost with the neuroplasm or from the vesicles during purification.
