Abstract

A simplified method for the chemical determination of total ketone bodies which eliminates autoclaving, distillation and refluxing is described. Acetoacetate and 3-hydroxybutyrate are converted to acetone and the latter is determined colorimetrically asacetone-2,4-dinitrophenylhydrazone. The method is highly sensitive and reproducible. Recoveries of individual ketones are quantitative. A series of normal values for total ketone bodies in whole blood, cerebrospinal fluid and 24-h urine is reported. Blood and urine ketone levels determined in fasting obese subjects are presented to illustrate the application of this method to ketosis.
