Abstract

In the current study, De Smet et al (J Clin Invest 2021;131:e137752, PMID 33621213) investigated the roles of connexin43 (Cx43), a major cardiac connexin, as hemichannels. Specifically, the authors determined the conditions underlying Cx43 hemichannel activation in the intercalated disc in ventricular cardiomyocytes, and their contribution to diastolic Ca2+ release from the sarcoplasmic reticulum and to electrical stability. The investigators demonstrated that Cx43 hemichannels were activated during diastolic Ca2+ release in single ventricular cardiomyocytes and cardiomyocyte cell pairs.
